In most cases, but not always. An infield single for instance may score a runner from third, but not from second. A runner can also score from third on a passed ball or wild pitch.

Upon Edit: A balk is another play where the extra base would make a big difference. I believe Armando balked in Reyes from third a few years back.

Santana will learn that he needs to go the distance and do the job himself if he wants to get a win. The Mets bullpen just can't be trusted to hold a lead and preserve a win.
